Omeprazole Pellets

Omeprazole pellets are enteric-coated granules containing omeprazole, a proton pump inhibitor that reduces stomach acid production. The coating protects the drug from stomach acid and ensures release in the intestine, making them effective for treating conditions like acidity, GERD, and ulcers.

Description

Omeprazole pellets are small, free-flowing spherical granules containing the proton pump inhibitor (PPI) omeprazole, used primarily for reducing gastric acid secretion. These pellets are typically enteric-coated to protect the drug from degradation in the acidic environment of the stomach and to ensure release in the intestine.

Mechanism of Action: Omeprazole irreversibly inhibits the H⁺/K⁺-ATPase enzyme system (proton pump) in gastric parietal cells, leading to a significant reduction in stomach acid production.

Formulation: Pellets are usually prepared by layering omeprazole onto inert cores (like sugar spheres) followed by an enteric coating. This coating dissolves at higher intestinal pH (≥5.5–6), enabling targeted drug release.

Uses: Widely used in the treatment of conditions such as:

  • Gastroesophageal reflux disease (GERD)
  • Peptic ulcer disease
  • Zollinger–Ellison syndrome

Advantages of Pellet Form:

  • Improved stability of acid-labile omeprazole
  • Uniform distribution in GI tract
  • Flexible dosing (can be filled into capsules or sachets)
  • Reduced risk of dose dumping compared to single-unit dosage forms

Administration: Usually filled into hard gelatin capsules or compressed into tablets. Pellets can sometimes be opened and sprinkled on soft food without crushing.
