Description
Trimix Injection is a compounded, sterile injectable medication used for the treatment of erectile dysfunction (ED) in adult males who have not responded adequately to oral PDE5 inhibitors (e.g., sildenafil, tadalafil). It contains a synergistic combination of three vasoactive agents designed to induce a firm and sustained erection through local action on penile tissue.
- Alprostadil (Prostaglandin E1) – 5 to 40 mcg/mL: Directly stimulates adenylate cyclase, increasing cAMP and promoting smooth muscle relaxation in the corpus cavernosum. Also known for independent erection induction without sexual stimulation.
- Papaverine Hydrochloride – 15 to 60 mg/mL: A non-selective phosphodiesterase inhibitor that relaxes vascular smooth muscle, leading to vasodilation.
- Phentolamine Mesylate – 0.5 to 2 mg/mL: An alpha-adrenergic antagonist that blocks sympathetic tone, preventing vasoconstriction and enhancing arterial inflow.
Mechanism of Action: When injected directly into the corpus cavernosum, the components of Trimix act synergistically to:
- Relax corporal smooth muscl
- Dilate cavernosal arteries
- Enhance arterial blood inflow
- Reduce venous outflow via veno-occlusion
This results in a rigid penile erection that typically occurs without the need for sexual stimulation, making it effective in cases of neurogenic or vasculogenic ED.
